Has anyone looked seriously at the relationship between time between races and pool size...and what an optimum time might be?

You would imagine that tracks have studied this.
They have to consider horsemen and the pari-mutuel office.

I don't think it's physically possible to run races with less than a 20 minute gap between them, added to the fact that pool size deteriorates if you don't allow enough time to bet.

Optimal time, I would guess, is somewhere around 25-30 minutes between races. JMO, though.
